I'm from New England but I was out in Chicago back in the 90's and we went to Kingston Mines on a Saturday night and it was an incredible experience. First of all, I wasn't into the blues back then as I am now so I don't even remember who was playing. What I remember, however, was the awesome setup. The bar was split in two with two separate rooms. Two bands set up, one in each room. One band would do a set, then take a break. All the patrons would get up and move to the other room and watch the second band jam for a while. This went back and forth all night. I think there were 4 bands in total that played. If I ever get back to Chicago Kingston Mines is a priority for me.

Its not a blues club, but if you're a harp player, stop by the Old Town School of Folk Music on a Monday night. That's the night for the group harp lessons, and the players are incredible and incredibly nice. They quit around 10:00, then head to a local bar to shoot the shit.
