I think it is just part of Cotton's style to play a note or two bent a little extra on 2 and 3 note chords. He doesn't play by the way it sounds, but by the way it feels. The video below is a great example as he does it several times...and I love the way at 1:08 that he takes the two hole draw from the deepest bend all the way up and everything in between. James Cotton is one of my favorites. Probabaly one of the most soulful players ever. I can always pick out when it is him backing on a Muddy Water's track.
In that video, that harp is tuned to 7LJI, which means certain notes of the chord are flatter but certain notes are also sharper as well so it produces a much truer harmony than ET does.
